Should I finish reading the Grisha trilogy before reading Six Of Crows (when it comes out, of course)? Or, are they two completely separate story lines?

Leigh Bardugo Six of Crows takes place two years after the events in Ruin and Rising so it will spoil some of the plot points in that book for you. If that's something you care about, finish the trilogy first.

BUT you definitely don't need to read the trilogy to understand what's happening in Six of Crows.
