The first thing you should do is get a professional editor. I'd recommend Caryn from I'm The Write Girl. (http://imthewritegirl.com/) I use her for all of my stuff, and she can give you tips for improving big picture stuff, as well as the nitty-gritty grammar. Then, find yourself a crit partner you really trust. Ideally, it should be someone who enjoys the genre in which you write, and who will have no problem being completely honest with you. Preferably someone you already know.
They're the main things I'd advise you to do at this stage. If your manuscript is still your first draft, I'd say EDIT, EDIT, EDIT. Everyone's different, but I need to revise my whole draft at least 4 or 5 times before I'm happy with it. I see the 1st draft as a lump of coal which editing gradually turns into a diamond.
